Endothelial Nitric Oxide Synthase (eNOS) and physiology cardiovascular system

Abstract:
Endothelial Nitric Oxide Synthase (eNOS) is essential for cardiovascular physiology due to its ability to modulate the function of blood vessels through the synthesis of NO. This review also discusses other roles of eNOS in cardio-vascular health besides the central role of regulating the vascular tone of blood vessels and health of endothelial cells which forms the basis of cardio-vascular diseases. In recently, eNOS is involve in the regulation of blood flow, prevention of platelet aggregation and maintaining the integrity of endothelial cells. Dysfunctional eNOS associated with impaired NO synthesis including the one that generates toxic superoxide anions is implicated in hypertension, atherosclerosis and stroke. This review discusses the structural and the biochemical aspects of eNOS focusing on the enzymatic function of eNOS, the post-translational modifications of eNOS and the regulation of eNOS. It also considers the effect of genetic factor and environmental factors on the regulation and function of eNOS; the paper shows that genetic variations and oxidative stress all significantly impact the function of eNOS and the progression of disease. In this regard, the present review has sought to highlight these aspects so as to enhance the understanding of the significance of eNOS in cardiovascular physiology and pathophysiology as well as to possibly define therapeutic targets for ameliorating cardiovascular prognosis. Further research agenda in this area involves investigating the factors behind the dysfunction of eNOS at the molecular level and discovering effective therapeutics to improve it in the hope of future prevention and treatment of cardiovascular ailments. For this reason, such dynamics have to be grasped in an effort to develop measures that can help curb the eNOS related dysfunction while encouraging the tickness of cardiovascular health achievement.
