Acute toxicity signs, lethal dose determination of Chalcone and its analogues in mice

Abstract:
Introduction: Toxicity profile is necessary for the approval of drugs and food products for human and animal consumption; this is to ensure the safety of the drug or food products. Acute toxicity is the toxicity that occurs after a single or multiple dose administration over a period of 24 hrs while LD50 is the dose of the drug that will kill 50% of the test animal population. Chalcone is a chemical compound consisting of two aromatic rings joined by α, β-ketone molecule that are not saturated with various substituents on the two aromatic rings. They are used as antimicrobial, anticancer among others. The study is targeted to determine acute toxicity signs and LD50 of chalcone and its derivatives.
Method: The samples, eight (8) derivatives of chalcone, or benzylideneacetophenone, were appraised for acute toxicity signs and LD50 using the Lorke's method. Twelve (12) mice were used for each derivative, and for the phase one (I): nine mice were divided into three (3) groups of three (3) mice while in the phase two (II): three (3) mice were divided into three groups with a mouse per group. The derivatives were administered orally and the animals observed for mortality and acute toxicity signs for 24 hrs.
Result: The result showed some derivatives of chalcone; benzylideneacetophenone displayed some toxicity signs, mortality and others mild toxicity signs, without mortality.
Conclusion: Two (2) of the eight (8) derivatives were more toxic with an LD50 of 3807.9 mg/kg compared to the other derivatives whose LD50 were > 5000 mg/kg.
