Optimization of nutritional and physiological conditions for antibiotic production by Endophytic Bacilus subtillis

Abstract:
In the present investigation optimum nutritional and physiological conditions for antibiotic compounds production by potential root endophytic bacterial isolate CREB-37 (B. subtilis) were assessed. To determine the best carbon and nitrogen sources for cell growth and antibiotic production, the endophytic antagonistic bacterial isolate was grown on medium containing different carbon sources (Mannitol, Glycerol and Xylose) and nitrogen sources (Soytone, Peptone and Malt extract) for 48 hrs. For determination of optimal nutritional concentrations, 0 % to 2.5 % of the selected carbon (Mannitol) and nitrogen (Soytone) sources was used. The culture temperature and initial pH for maximum growth and production of antibiotic substances from antagonistic microorganism was determined by incubating for 48 hrs at 20 to 40 oC and 4.0 to 10.0 respectively. Mannitol @ 2 %, soytone @ 2 %, pH 7 and temperature 30 oC were found to be optimum for cell growth and antibiotic production. Similarly, significant improvement in the cell growth and antibiotic production was observed with the modified Luria broth than the basal medium (Luria broth).
