β - Galactosidase assay on microfluidic paper-based analytical devices (µPADs)

Abstract:
Enzyme assays using Microfluidic Paper-Based Analytical Devices (µPADs) are effective yet economical alternatives to conventional tube based enzyme assays. The current work involves standardisation of β-Galactosidase (β-gal) assay on µPADs using Chlorophenol red-β-D-galactopyranoside (CPRG) as a substrate. The assay was standardised with respect to substrate concentration, time of incubation, working range of enzyme and the limit of detection on the device. Presence of β-gal producers (coliforms) in spiked food samples using this assay was demonstrated as a proof of concept.
